We were awarded a 5 year NIH/NIGMS grant to study the mechanisms of coordination of transcription and DNA replication in eukaryotic cells! The whole lab have contributed to generation of exciting preliminary data that promise great findings in the near future. Stay tuned for job opportunities!

Today we welcomed OMRF Fleming Scholars Sophia and Fatima in the lab. They will work on the NSF-funded project focused on the roles of conserved domains of BET proteins in transcription regulation.

A new manuscript from the lab is on bioRxiv! We evaluated the importance of all conserved domains of BET proteins and determined that the extra-terminal (ET) domain is essential for cell viability, genome-wide transcription and BET protein chromatin occupancy. Furthermore, we provided evidence that the ET domain performs these functions serving as a hub for interactions with other factors involved in transcription regulation. Specifically, we showed that the ET domain recruits coactivators TFIID and Mediator to promoters and revealed mechanistic basis for a previously reported Bdf1-TFIID interaction by identifying and validating the ET-interacting motifs on a TFIID subunit Taf7. Our findings expand current understanding of the complex biology of BET proteins and suggest the mechanisms by which cells can bypass bromodomain inhibition in pathologic states.

Iwona joined us today for a year-long traineeship. Iwona is finishing her Master's degree in Molecular Biotechnology at the Jagiellonian University in Poland. She will continue a project started by Agnieszka that focuses on a functional dissection of the mechanisms of BET proteins in transcription regulation.

We posted our first manuscript on bioRxiv. We describe a step-by-step protocol combining rapid degradation of target proteins using AID system with quantitation of newly synthesized mRNA using SLAM-seq. We also provide an extra-cheap protocol for library preparation for less than $15 per sample. The SLAM-seq and library preparation protocols we verified both in yeast and in mammalian cell line models.
